Cover: Expression of aquaporin-3 in human proximal bronchioles. Aquaporin-3 is the only aquaporin found expressed in the human bronchioles, and has a high level of expression in the superficial epithelium of proximal (pseudostratified) bronchioles. Upper panels show a human bronchiole in cross section. Left, in situ hybridization with an anti-sense human aquaporin-3 probe showing intense signal detected throughout the epithelium. Right, bright field H&E stain of the same field as shown in the left panel (magnification ×80). Lower panels are representative immunofluorescence analyses of the same bronchiolar tissue with aquaporin-3 antibodies (left) and non-immune rabbit IgG (right). Both sections were exposed to Texas Red labeled anti-rabbit secondary antibody and were counterstained with DAPI. Aquaporin-3 antibodies specifically stain the apical membrane of columnar cells, as well as the plasma membrane of basal cells (magnification ×600, confocal xy scanning, z ~ 1 µm).
